Los 265 jugadores elegibles para el draft consisten en 175 jugadores que obtuvieron elegibilidad a través de la NBA 2K League Draft Prospect Series, 22 jugadores de eventos clasificatorios internacionales en las últimas dos temporadas bajas, cinco jugadores de la iniciativa Women in Gaming de la NBA 2K League, cinco jugadores que fueron identificados por la NBA 2K League y sus equipos como jugadores de alto rendimiento en la comunidad 2K, y 58 jugadores no retenidos de la temporada 2020.
